The experiments are designed to determine the conditions required for the initiation of allograft rejection and to examine the nature of the cells that are necessary to carry out this process. Further, the possibility that allograft rejection is mediated by class II restricted, epitope specific, cytotoxic T cells will be explored. We will also examine the induction of tolerance by donor corneas in allosensitized mice, and determine the role of class I MHC molecules in this process. Finally, we will begin to characterize the molecular events that occur in corneal endothelial cells that lead to activation of allospecific T cells.
